"There are no words to convey my shock and sadness regarding the tragic accident that took the life of Halyna Hutchins, a wife, mother and deeply admired colleague of ours," he wrote in a series of posts on Twitter.
"I am in touch with her husband, offering my support to him and his family.
"My heart is broken for her husband, their son, and all who knew and love Halyna."
Photos showed the 68-year-old actor hunched over, upset and distressed moments after the incident.
According to witness reports, Baldwin asked: "Why was I handed a hot gun?"
Hutchins, 42, the director of photography for the film, died when Baldwin fired a prop gun reportedly filled with blanks that also injured 48-year-old director Joel Souza.
Hutchins was flown to the University of New Mexico Hospital where she was pronounced dead. Mr Souza was taken by ambulance to Christus St Vincent's Hospital and has since been released, according to a representative.
Her grieving husband, Matthew, has told publication Daily Mail, Baldwin had been in contact with him and was "being very supportive".
A statement from the Santa Fe County Sheriff’s office said the pair “were shot when a prop firearm was discharged by Alec Baldwin, 68, producer and actor”.
"According to investigators it appears the scene being filmed involved the use of a prop firearm when it was discharged."
"Detectives are investigating how and what type of projectile was discharged."
Baldwin said he will be "fully cooperating" with the police investigation to find out how the incident occurred.
Executive director Rebecca Rhine and President of the International Cinematographers Guild, Local 600, John Lindley said in a joint statement they would support an investigation into the incident.
“We received the devastating news this evening, that one of our members, Halyna Hutchins, the Director of Photography on a production called ‘Rust’ in New Mexico died from injuries sustained on the set,” the statement read.
“The details are unclear at this moment, but we are working to learn more, and we support a full investigation into this tragic event. This is a terrible loss, and we mourn the passing of a member of our Guild’s family.”
Ukraine-born Hutchins was named a "rising star" by American Cinematographer in 2019 with colleagues describing the mother-of-one as a "genius".
Just two days earlier a smiling Halyna was pictured alongside Baldwin in a picture of the cast and crew on the movie set.
Baldwin is a co-producer and stars in the Western film as outlaw "Rust", who travels to Kansas to break his estranged 13-year-old grandson out of prison after he was convicted of an accidental murder and sentenced to hang.
Production of the film has been halted as investigations continue.
